从我国东北林区死者脑组织中,分离到蜱传脑炎病毒HLJ-1株,测定了它的E蛋白基因序列和推导的氨基酸序列,以及E蛋白抗原位点反应图谱。将该株与远东亚型Sofyn株和中欧亚型Neuderfl株做了同源性比较,HLJ-1株与Sofyn株E蛋白基因的核苷酸及氨基酸的同源性分别为94.3%和98.8%;与Neudoerfl的核苷酸和氨基酸的同源性分别为83.7%和96%,在E蛋白的核苷酸序列中,HLJ-1和Sofyn株之间存在着85个碱基的变异和6个氨基酸的改变,变化的氨基酸分别是Ser-438→Gly,Arg-451→Lys,Ala-612→Val,Met-638→Ile,Asn-646→Thr,Val-682→Ala。从E蛋白的模型得知,变异的氨基酸位于E蛋白的保守区域,故对抗原性影响不大。另E蛋白位点总图谱显示,HLJ-1与Sofyn株在C位点的反应存在差异,这可能是由于培养液低pH引起HLJ-1的空间结构改变所致。本文首次从基因水平证实了从东北林区分离的蜱传脑炎病毒属于远东型蜱传脑炎病毒。
A strain of tick-borne encephalitis virus HLJ-1 was isolated from the brain tissue of deceased persons in the northeastern China. The E protein gene sequence and deduced amino acid sequence and the E protein antigen site reaction map were determined. The homology comparison between this strain and the Sofyn strain of Far East Asia and the Neuderfl strain of Central Europe was carried out. The nucleotide and amino acid homologies of the E protein genes of HLJ-1 and Sofyn strains were 94.3% and 98.8%. The nucleotide and amino acid identities with Neudoerfl were 83.7% and 96%, respectively. In the nucleotide sequence of E protein, there were 85 bases between HLJ-1 and Sofyn strains Amino acid changes Ser-438 → Gly, Arg-451 → Lys, Ala-612 → Val, Met-638 → Ile, Asn-646 → Thr, Val-682 → Ala . According to the model of E protein, the variant amino acids are located in the conserved region of E protein and therefore have little effect on antigenicity. The other E site map showed that HLJ-1 and Sofyn strain at the C site reaction differences, which may be due to the low pH of the culture medium caused HLJ-1 spatial structure changes caused. This paper for the first time confirmed from the gene level that the tick-borne encephalitis virus isolated from the northeast forest belonged to the Far Eastern type tick-borne encephalitis virus.
