目的分析评价凝胶抗球蛋白试验(GAT)的特点,增加输血安全性。方法以不同稀释度的商品化抗D(IgG)为基础制备不完全抗体和致敏红细胞为模拟标本,通过一系列实验并结合文献分析GAT(DiaMed的LISS/Coombs卡)的特点。结果红细胞洗涤与否不影响GAT检测的结果;外加AB血浆至50μl后,才使GAT产生的强阳性略有减弱;GAT可测抗D(IgG)最高效价为1:256。结论GAT检测过程中不必洗涤红细胞,使用配套稀释液可提高检测灵敏度和准确性。GAT反应灵敏度明显高于试管法,而近似于聚凝胺法。
Objective To analyze and evaluate the characteristics of GAG in order to increase the safety of blood transfusion. METHODS: Incomplete antibody and sensitized erythrocytes were prepared on the basis of different dilutions of commercially available anti-D (IgG). The characteristics of GAT (DiaMed’s LISS / Coombs Card) were analyzed by a series of experiments and literature. Results The results of GAT test did not affect the results of GAT test. The strong positive of GAT was slightly weakened after addition of AB plasma to 50μl. The highest titer of GAT was 1: 256. Conclusion No need to wash red blood cells during the GAT test. The use of complementary diluents can improve the detection sensitivity and accuracy. GAT reaction sensitivity was significantly higher than the tube method, and similar to the polybrene method.
