自1994年Zhang等首次从小鼠和人的脂肪组织中分离并克隆出肥胖基因(ob基因)以来,关于肥胖发生的分子机制这一领域已成为举世瞩目的热点。肥胖基因编码的mRNA表达产物-肥胖蛋白Leptin的研究更是占据了肥胖研究的核心地位。Leptin是ob基因在脂肪组织表达的一种蛋白质,其受体主要存在于下丘脑,在其他部位如胰岛β细胞、肝、肺、心、肾、睾丸、脂肪组织等中也有Leptin受体表达,但水平较低。随着对Leptin研究的不断深入,人们发现Leptin具有广泛的生理功能。对Leptin的研究对揭示肥胖、糖尿病、高血压等多种疾病具有重要意义。笔者从Leptin的功能及作用机制方面作一综述。
Since the first time that Zhang et al. Isolated and cloned fat gene (ob gene) from mouse and human adipose tissue in 1994, the molecular mechanism of obesity has become a hot spot in the world. Obesity gene encoding mRNA expression product - obesity protein Leptin research is occupy the core of obesity research. Leptin is a protein expressed in adipose tissue of ob gene. Its receptor mainly exists in the hypothalamus. Leptin receptor is also expressed in other sites such as islet β cells, liver, lung, heart, kidney, testis, adipose tissue, But the level is lower. With the deepening of Leptin research, Leptin has been found to have a wide range of physiological functions. Research on Leptin is of great importance to reveal various diseases such as obesity, diabetes and hypertension. The author reviews the function and mechanism of Leptin.
