目的探讨不同地域间屋尘螨和粉尘螨主要变应原变异体的差异。方法分别提取中国华南地区培养的粉尘螨和欧洲商业化培养的屋尘螨和粉尘螨的螨体、螨卵和全螨培养物总RNA,采用RT-PCR技术扩增第1组变应原的包含前导肽段、酶原肽段和成熟肽段,以及第2组变应原的包含前导肽段和成熟肽段的目的片段,根据国际标准参照物进行序列比较。结果欧洲商业化的屋尘螨Derp1主要变异体是Derp1.0105,除了第124V、50Y和19M位氨基酸有频繁的置换外,其余为散在的置换,出现8种新的变异体;Derp2主要变异体是Derp2.0104,置换集中在第40V、4T7、111M和114D位,出现6种新的变异体。中国华南地区的粉尘螨Derf1变异体很少,出现3种新的变异体;与欧洲商业化的粉尘螨相比,中国华南地区有不同的Derf2变异体,出现6种新的变异体。来自螨体、螨卵和全螨培养物的各主要变应原变异体无明显差异。结论不同地域间尘螨的主要变异体不同。
Objective To explore the differences of major allergen variants between house dust mites and dust mites in different regions. Methods The total RNA of mite body, mite egg and total mite culture of house dust mites and house dust mites cultured in South China were extracted from China, respectively. The allergen of group 1 The target fragment containing the leader peptide, the proenzyme peptide and the mature peptide, and the second group of allergens, including the leader peptide and the mature peptide, were compared according to the international standard reference. Results Derp1.0105, a major commercial variant of Derp1 in commercialized house-dust mite, was found to be scattered in place of the 124V, 50Y and 19M amino acids. Eight new variants were found. Derp2 major variant Is Derp2.0104, with substitutions concentrated at positions 40V, 4T7, 111M and 114D, and 6 new variants appeared. There are only a few Derf1 variants of Dust mite in South China, and three new variants appear. Compared with European commercial Dust Mites, there are different Derf2 variants in southern China and six new variants appear. There was no significant difference in the major allergen variants from mites, mite eggs and whole mite cultures. Conclusion The main variants of dust mites in different regions are different.
