目的 :在人胎儿脑组织 c DNA中钓取并克隆新基因。方法 :以 RT- PCR方法用人 FGF- 19特异性引物从人胎脑 c DNA中扩增到一大小约 6 6 0 bp的片段 ,并连入 TA克隆载体测序 ,得到一个表达序列标签 (EST) ,输入 Gene Bank、Swissprot、db EST等数据库中进行同源比较分析。结果 :分析表明它是人胎儿脑组织未知基因 EST,是一个 6 6 3个碱基可编码 2 2 0个氨基酸的开放读码框架。结论 :在人胎儿脑组织 c DNA中钓取并克隆一个人胎脑新基因 EST。
OBJECTIVE: To fetch and clone new genes in c DNA of human fetal brain tissue. Methods: A fragment of about 6 60 bp in length was amplified from human fetal brain cDNA by RT-PCR using human FGF-19-specific primers and sequenced to obtain a sequence tag (EST) , Enter Gene Bank, Swissprot, db EST database for comparative analysis of homology. Results: The analysis showed that it is an unknown human fetal brain gene EST, which is an open reading frame of 6 6 3 bases encoding 220 amino acids. Conclusion: A human fetal brain new gene EST was captured and cloned in c DNA of human fetal brain tissue.
