目的建立小菜蛾抗菌肽CA的高效液相色谱测定方法。方法色谱柱为Agilent Alltima C18(4.6 mm×250 mm,5μm),流动相为甲醇-乙腈-0.05%三氟乙酸溶液(2.5∶2.5∶95),检测波长为275 nm,柱温为25℃,流速为1.0 mL·min-1。结果小菜蛾抗菌肽CA的线性范围为0.08~0.800μg(r=0.999 4,n=6),平均回收率分别为99.69%,RSD为1.20%(n=6)。结论本法快速、简便、准确,重现性好,可用于小菜蛾抗菌肽CA的定量分析和质量控制。
OBJECTIVE To establish a method for the determination of antimicrobial peptide CA in Plutella xylostella by high performance liquid chromatography. METHODS The column was Agilent Alltima C18 (4.6 mm × 250 mm, 5 μm) with a mobile phase of methanol-acetonitrile-0.05% trifluoroacetic acid (2.5: 2.5: 95) at a detection wavelength of 275 nm and a column temperature of 25 ° C. The flow rate was 1.0 mL · min-1. Results The linear range of antibacterial peptide CA of Plutella xylostella was 0.08-0.800 μg (r = 0.999 4, n = 6). The average recovery was 99.69% and the RSD was 1.20% (n = 6). Conclusion The method is rapid, simple, accurate and reproducible. It can be used for the quantitative analysis and quality control of antibacterial peptide CA in Plutella xylostella.
