目的:探讨血清反应因子(SRF)在肾小管上皮细胞间充质转分化(EMT)过程中的作用和可能机制。方法:将54只清洁级(SPF级)SD大鼠中36只行单侧输尿管结扎(UUO组),另外18只为假手术组(Sham组)。在UUO术后3 d和14 d分别处死18只大鼠并留取肾脏组织,应用免疫组织化学方法检测肾小管上皮细胞中SRF、E-cadherin、α-SMA和Snail的表达变化。结果:UUO组3 d和14 d的肾组织中SRF、α-SMA和Snail表达显著增高(P<0.05),其中SRF在3 d和14 d的阳性率分别为83.3%、94.4%,假手术组为5.56%;E-cadherin表达显著下降(P<0.05)。结论:SRF可能参与了单侧输尿管结扎大鼠肾小管上皮间充质细胞转分化的过程。
Objective: To investigate the role of serum factor (SRF) in the process of renal tubular epithelial mesenchymal transition (EMT) and its possible mechanism. Methods: Thirty - six clean - grade (SPF) SD rats were unilateral ureteral ligation (UUO group) and the other 18 sham group (sham group). Eighteen rats were sacrificed on the third and fourth postoperative day of UUO, and the kidneys were removed. The expressions of SRF, E-cadherin, alpha-SMA and Snail in renal tubular epithelial cells were detected by immunohistochemistry. Results: The expression of SRF, α-SMA and Snail in 3 and 14 d of UUO group were significantly increased (P <0.05). The positive rates of SRF on 3 d and 14 d were 83.3% and 94.4% Group was 5.56%; E-cadherin expression was significantly decreased (P <0.05). Conclusion: SRF may be involved in the process of renal tubular epithelial mesenchymal cells transdifferentiation in unilateral ureteral obstruction rats.
