把BEL-7402细胞株成功地移植于裸鼠,并已传5代。观测瘤体的生长情况,从生长曲线表现出肿瘤有较高的恶性行为——不断膨胀性生长。移植瘤病理形态学上与原细胞株具有相似性,移植瘤细胞核大,细胞多形性,细胞排列有的呈索状或腺样。电镜下移植瘤细胞具肝癌细胞超微结构特征,如细胞之间形成发育不全毛细胆管及其细胞连接,此外,还见到胞浆内存在核膜组装过剩的环孔板、染色质间颗粒、张力原纤维等结构。移植瘤染色体众数为不足三倍体,与原细胞株染色体众数分布大体相同。应用抗7402细胞多抗,PAP染色法表明移植细胞阳性反应,胞浆内有棕褐色沉淀。实验过程中移植瘤未测得AFP。这与原细胞株上清AFP阴性一致,移植瘤是否会出现AFP阳性反应有待于继续观测。
The BEL-7402 cell line was successfully transplanted in nude mice and passed on for 5 passages. The number of xenograft chromosomes is less than triploid, which is roughly the same as that of the original cell line. Application of anti-7402 cell polyclonal antibody, PAP staining showed that transplanted cells were positive, cytoplasm with brown precipitate. AFP was not detected in the xenografts during the experiment. This is consistent with the original cell line AFP negative supernatant, transplanted tumor will appear AFP positive reaction to be observed.
