目的:检测黑素瘤抑制蛋白(MIA/CD-RAP)在小鼠胚胎乳腺组织中的表达。方法:剖腹分离出10.5~16.5 d 的MIA/CD-RAP启动子-半乳糖苷酶(2.2lacZ)转基因小鼠胚胎,利用X-gal全胚胎染色观察转基因的组织和细胞染色,并以免疫组织化学染色检测内源性MIA/CD-RAP蛋白在胚胎乳腺组织中的表达。结果:MIA/CD-RAP在孕期11.5 d 的小鼠胚胎乳腺上皮细胞中开始表达,至13.5 d 表达明显增强,而后逐渐减弱,至16.5 d 消失。结论:MIA/CD-RAP可能在乳腺的早期发育和乳腺癌发生中起重要作用。
Objective: To detect the expression of melanoma inhibitor protein (MIA / CD-RAP) in mouse embryonic breast tissue. METHODS: MIA / CD-RAP promoter-galactosidase (2.2lacZ) transgenic mouse embryos were isolated from 10.5-16.5 d by Caesarean section. Tissue and cell staining of transgenic tissues were observed by X-gal whole embryo staining , And the expression of endogenous MIA / CD-RAP protein in embryonic breast tissue was detected by immunohistochemical staining. Results: MIA / CD-RAP began to express in mouse embryonic mammary epithelial cells at 11.5 days of gestation. The expression of MIA / CD-RAP was significantly enhanced at 13.5 days and then gradually weakened and disappeared after 16.5 days. Conclusion: MIA / CD-RAP may play an important role in the early development of mammary gland and in the development of breast cancer.
