目的评估线性探针方法快速检测结核分枝杆菌的耐药性效果。方法选取2013年-2014年江西省耐药监测点分离培养的阳性菌株,收集菌体,提取DNA,采用MTBDRplus试剂盒进行检测,以国际标准的比例法检测结果作为对照,评价其敏感度及特异度,并分析突变分布情况。结果检测利福平耐药的灵敏度为93.33%,特异度为98.35%;检测异烟肼的灵敏度为76.31%,特异度为98.83%。耐利福平基因突变频率最高的位点为rpo B H531L(57.89%),耐异烟肼基因突变频率最高的位点是kat G S315T1(60.61%)。结论可利用MTBDRplus快速检测利福平和异烟肼的耐药性,是快速发现耐药结核分枝杆菌的有效方法。
Objective To assess the drug resistance of Mycobacterium tuberculosis by linear probe method. Methods The positive strains isolated from drug-resistant surveillance sites in Jiangxi Province from 2013 to 2014 were selected and the cells were harvested for DNA extraction. MTBDRplus kit was used to detect the positive strains. The sensitivity and specificity Degree, and analyze the distribution of mutations. Results The sensitivity of detecting rifampicin was 93.33% and the specificity was 98.35%. The sensitivity and specificity of isoniazid were 76.31% and 98.83% respectively. The most frequent site of rifampicin gene mutation was rpo B H531L (57.89%). The site with the highest frequency of isoniazid resistance mutation was kat G S315T1 (60.61%). Conclusion MTBDRplus can be used to rapidly detect the drug resistance of rifampicin and isoniazid, and it is an effective method to rapidly detect drug-resistant Mycobacterium tuberculosis.
