目的 :探讨不同组织病理分级的星形细胞瘤中PTEN、Mdm2和p5 3的表达水平 ,并分析PTEN影响Mdm2、p5 3表达的信号转导机制。方法 :采用免疫组织化学方法检测 68例星形细胞瘤标本中PTEN、Mdm2和p5 3的表达水平。结果 :星形细胞瘤中PTEN、Mdm2、p5 3的表达水平分别是 5 4.4% (3 7/ 68)、41.2 % (2 8/ 68)、45 .6% (3 1/ 68)。PTEN阳性标本中Mdm2的表达率 (2 4.3 % ,9/ 3 7)与PTEN阴性标本中该蛋白的表达率 (61.3 % ,19/ 3 1)相比差异显著 ,统计学分析显示PTEN表达与Mdm2表达呈负相关 (P <0 .0 1)。Mdm2表达和p5 3表达一致符合率达 66.2 % (45 / 68) ,两者的表达密切相关 (P <0 .0 5 )。结论 :(1)PTEN、Mdm2和p5 3表达与星形细胞瘤的组织病理分级相关。 (2 )抑癌基因PTEN可以下调癌基因Mdm2的表达水平。 (3 )Mdm2和p5 3的表达存在一致性
Objective: To investigate the expression of PTEN, Mdm2 and p5 3 in different histopathological grades of astrocytomas and to analyze the signal transduction mechanism of PTEN on the expression of Mdm2 and p5 3. Methods: Immunohistochemistry was used to detect the expression of PTEN, Mdm2 and p5 3 in 68 specimens of astrocytomas. Results: The expression of PTEN, Mdm2 and p5 3 in astrocytomas was 54.4% (37/68), 41.2% (28/68) and 45.6% (31/68), respectively. The expression of Mdm2 in PTEN positive specimens was significantly higher than that in PTEN negative specimens (61.3%, 19/3 1). The statistical analysis showed that the expression of Mdm2 was positively correlated with Mdm2 Expression was negatively correlated (P <0.01). The consistent rate of Mdm2 expression and p5 3 expression was 66.2% (45/68), which was closely related to the expression of p5 3 (P <0.05). Conclusion: (1) The expression of PTEN, Mdm2 and p5 3 are related to the histopathological grade of astrocytoma. (2) The tumor suppressor gene PTEN can down-regulate the expression level of oncogene Mdm2. (3) The expression of Mdm2 and p5 3 are consistent
