胚胎发育晚期蛋白是植物在逆境胁迫下产生的一种应激蛋白质,具有较高的亲水性和热稳定性,与植物抗逆功能密切相关.本研究从己构建的柠条锦鸡儿干旱胁迫抑制削减杂交文库(SSH)中筛选到一条LEA蛋白编码基因的部分序列,用RACE技术扩增得到该基因cDNA全长.序列比对与系统进化分析显示,该基因没有特定结构域,它与拟南芥LEA4 (AT3G53040)基因同源性最近,故命名为CkLEA 4-3.该基因cDNA长971 bp,开放阅读框架长636 bp,编码21 1个氨基酸,推测蛋白分子量为22.44kD,等电点为5.76,是一种亲水蛋白.利用荧光定量PCR技术检测发现,CkLEA 4-3基因在干旱、ABA、NaC1和脱水处理下均受到不同程度的诱导,推测CkLEA 4-3基因可能与柠条锦鸡儿响应逆境胁迫有关.“,”Late embryogenesis abundant (LEA) is a stress protein produced by plants under stress conditions.It has a high hydrophilicity and thermal stability and plant resistance is closely related to function.In this study,parts of sequence in an LEA encoding gene fragment was isolated from the suppression subtractive hybridization library of Caragana korshinskii,and the full length eDNA was cloned by rapid amplification of cDNA ends technique.Sequence and phylogenetic analysis indicated that it had no specific domain.It was closest to Arabidopsis thaliana LEA 4 (AT3G53040) gene homology,so named CkLEA 4-3.Sequencing results showed that the full length eDNA was 971 bp,with an open reading frame of 636 bp.It encoded 211 amino acids with a calculated molecular mass of 22.44 kD,and a theoretical pI of 5.76.The deduced protein was strongly hydrophilic.Real-time quantitative PCR analysis showed that the transcript of CkLEA4-3 was induced in various degrees under different treatments,including ABA,dehydration,salt and drought.These results indicated that CkLEA 4-3 might be involved in stress responses ofC.korshinskii.
