目的:比较不同诱导方法对LAN细胞杀伤活性的影响。方法:外周血单个核细胞(PBMC)用重组白细胞介素-2(rIL-2)体外诱导LAK细胞,采用3H-TdR释放法测定LAK活性。结果;(1)PBMC诱导3h后,LAK活性高于或至少不低于24~72h者;(2)PBMC诱导3h后洗去游离的rIL-2,并不影响继续诱导LAN细胞活性;(3)PBMC分离后以清洗2次为佳;(4)PBMC用正常人血浆培养比用肝炎病人自体血浆培养诱导的LAK活性高。结论:在本试验系统中,PBMC分离后,清洗2次,以正常人血浆经IL-2诱导3h为最佳回输条件。
Objective: To compare the effects of different induction methods on the killing activity of LAN cells. Methods: Peripheral blood mononuclear cells (PBMCs) were induced by recombinant interleukin-2 (rIL-2) in vitro. LAK activity was measured by 3H-TdR release assay. Results: (1) After three hours of PBMC induction, the activity of LAK was higher or at least not lower than 24-72h. (2) The rIL-2 was washed away after 3h PBMC induction, ) PBMC after cleaning to wash 2 times is better; (4) PBMC with normal human plasma culture than with hepatitis patients autologous plasma culture induced LAK activity is high. CONCLUSION: In this test system, PBMCs were washed twice after separation, and the optimal conditions for reinfusion were as follows: normal human plasma induced by IL-2 for 3 hours.
