目的 :探讨Bax基因在热诱导BcaCD885细胞凋亡过程中的作用。方法 :水浴加温诱导BcaCD885细胞产生凋亡。通过FCM DNA、FCM 抗体检测及RT PCR技术 ,观测热诱导颊癌细胞凋亡过程中Bax基因蛋白及mRNA表达的动态变化。结果 :凋亡率与Bax蛋白表达水平存在正相关关系 ;加热后 ,细胞BaxmRNA相对量明显升高。结论 :高温作为一种外来刺激信号通过上调Bax基因的表达 ,诱导BcaCD885细胞凋亡的发生。
Objective: To investigate the role of Bax gene in heat-induced apoptosis of BcaCD885 cells. Methods: BcaCD885 cells were induced to apoptosis by water bath warming. FCM DNA, FCM antibody detection and RT PCR were used to observe the dynamic changes of Bax gene protein and mRNA expression in heat-induced apoptosis of buccal cancer cells. Results: There was a positive correlation between the apoptosis rate and the expression of Bax protein. After heating, the relative amount of Bax mRNA was significantly increased. Conclusion: High temperature as an external stimulus can induce the apoptosis of BcaCD885 cells by up-regulating Bax gene expression.
