目的对采自四川省凉山州会东县的白肉灵芝进行种属鉴定及抗氧化功效评价。方法基于样本的ITS序列完成系统发育分析,采用H_2O_2处理PC12细胞建模,MTT法检测细胞存活率,验证白肉灵芝水提物对PC12细胞的保护作用。结果样本与白肉灵芝Ganoderma leucocontextum同属于一个分支,相似度98%。样本水提物能显著提高H_2O_2处理后的PC12细胞的存活率并降低Caspase-3活性。结论样本鉴定为白肉灵芝Ganoderma leucocontextum,对H_2O_2诱导的PC12细胞凋亡有保护作用,其机制可能与抑制细胞凋亡线粒体途径中Caspase-3的表达有关。
Objective To evaluate the species identification and anti-oxidative efficacy of Ganoderma lucidum collected from Huidong County, Liangshan Prefecture, Sichuan Province. METHODS: Phylogenetic analysis was performed based on the ITS sequence of the samples. H 2 O 2 -treated PC12 cells were used for modeling. Cell viability was detected by MTT assay to verify the protective effect of Ganoderma lucidum aqueous extracts on PC12 cells. The results of samples and white Ganoderma Ganoderma leucocontextum belong to the same branch, the similarity of 98%. The sample water extract can significantly improve the survival rate of PC12 cells treated with H 2 O 2 and decrease the activity of Caspase-3. Conclusion The sample was identified as Ganoderma leucocontextum, which has a protective effect on the apoptosis of PC12 cells induced by H_2O_2. The mechanism may be related to the inhibition of the expression of Caspase-3 in mitochondrial pathway.
