目的:探讨苦参碱对人白血病HL-60细胞的生长抑制作用及对凋亡相关基因表达的影响。方法:HL-60细胞分为对照组、不同浓度苦参碱药物处理组和阳性对照组顺铂。采用MTT比色分析法检测苦参碱对HL-60细胞生长的抑制作用,采用TUNEL法测定细胞凋亡、比色法测定Caspase-3活性、实时定量RT-PCR法检测Bax、BCl-2、Caspase-9及Survivin的mRNA的表达。结果:苦参碱0.5 mg/ml组、1 mg/ml组和2 mg/ml组均能抑制HL-60细胞生长,其抑制作用呈浓度依赖性。苦参碱可诱导人白血病HL-60细胞凋亡并上调Caspase-3酶活性,同时苦参碱可减少BCl-2、Survivin的mRNA表达,增加Bax、Caspase-9的mRNA表达。结论:苦参碱可有效诱导HL-60细胞发生凋亡,其机制可能与上调Bax、Caspase-9的mRNA表达并抑制BCl-2、Survivin mRNA表达相关。
Objective: To investigate the inhibitory effect of matrine on the growth of human leukemia HL-60 cells and its effect on the expression of apoptosis related genes. Methods: HL-60 cells were divided into control group, matrine group and cisplatin group. MTT colorimetric assay was used to detect the inhibitory effect of matrine on the growth of HL-60 cells. The apoptosis of HL-60 cells was detected by TUNEL assay. The activity of Caspase-3 was determined by colorimetric assay. The Bax, Bcl- Caspase-9 and Survivin mRNA expression. Results: Matrine 0.5 mg / ml, 1 mg / ml and 2 mg / ml could inhibit the growth of HL-60 cells in a concentration-dependent manner. Matrine can induce the apoptosis of human leukemia HL-60 cells and up-regulate the activity of Caspase-3. At the same time, matrine can reduce the mRNA expression of Bcl-2 and Survivin and increase the mRNA expression of Bax and Caspase-9. Conclusion: Matrine can effectively induce the apoptosis of HL-60 cells. The mechanism may be related to up-regulating the mRNA expression of Bax and Caspase-9 and inhibiting the expression of Bcl-2 and Survivin mRNA.
