目的 :观察硫酸长春新碱 (VCR)控释药膜对动物实验中肿瘤的抑制作用及其急性毒性。方法 :VCR控释膜埋植入荷瘤小鼠体内 ,观察对小鼠艾氏实体瘤的抑制作用以及对血清SOD的影响 ;药膜于体外细胞培养液中进行释放 ,释放液同K562细胞共同培养 ,MTT法观察对K562细胞的抑制率 ,通过透射电子显微镜观察对K562细胞凋亡的影响。与静脉给药方式相比较 ,通过血液指标和病理组织切片观察其急性毒性。结果 :VCR控释药膜在体内、外均显著抑制肿瘤细胞的生长 ,增加SOD活性 ,并可诱导肿瘤细胞的凋亡发生 ,且毒性较静脉给药明显降低。结论 :VCR控释药膜可以有效地抑制肿瘤生长 ,并且毒性较低。
OBJECTIVE: To observe the inhibitory effect and acute toxicity of vincristine sulfate (VCR) controlled release film on tumor in animal experiments. Methods: VCR controlled release membrane was implanted into tumor-bearing mice to observe the inhibitory effect on Ehrlich solid tumors in mice and its effect on serum SOD. The membrane was released in vitro and the release solution was co-cultured with K562 cells The inhibitory rate of K562 cells was observed by MTT assay. The apoptosis of K562 cells was observed by transmission electron microscopy. Compared with intravenous administration, acute toxicity was observed by blood index and pathological section. Results: VCR controlled release film significantly inhibited the growth of tumor cells in vivo and in vitro, increased the activity of SOD and induced the apoptosis of tumor cells, and the toxicity was significantly lower than that of intravenous administration. Conclusion: VCR controlled release film can effectively inhibit tumor growth and has low toxicity.
