对31例脐血及31例正常成人骨髓细胞进行体外培养,分别检测其CFU-GM,BFU-E,巨核细胞集落形成单位(CFU-Meg),粒、红、巨噬细胞集落形成单位(CFU-GEM),粒、红、巨噬、巨核细胞集落形成单位(CFU-GEMM)及初始造血祖细胞——粘附于基质的原始细胞集落形成单位(sCFU-Bl),对比分析其造血细胞的层次结构。发现脐血晚期祖细胞含量接近骨髓,早期祖细胞含量则高于骨髓。经推算21天时sCFU-Bl含量可达骨髓的10倍左右。提示脐血长期重建造血的能力优于骨髓,一份脐血重建成人造血理论上是可能的。
Thirty-one cord blood samples and 31 normal adult bone marrow cells were cultured in vitro. The CFU-GM, BFU-E, CFU-Meg, granulocyte, red and macrophage colony forming units (CFU -GEM), granulocyte, erythrocyte, macrophage, megakaryocyte colony forming unit (CFU-GEMM) and the initial hematopoietic progenitor cells-sCFU-Bl adhered to the matrix, and compared their hematopoietic cells Hierarchy. Found that cord blood progenitor cells close to the bone marrow, the content of early progenitor cells is higher than the bone marrow. Calculated sCFU-Bl content of bone marrow up to 10 times 21 days after the projections. Tip cord blood long-term reconstruction of hematopoietic capacity better than the bone marrow, a cord blood reconstructed adult hematopoietic theoretically possible.
