目的研究卵巢切除后鼠椎间盘结构及软骨P物质神经分布变化,探索骨质疏松性腰背部疼痛的原因。方法3月龄雌性SD大鼠64只,分组行双侧卵巢切除(OVX)及假手术(Sham-O);术后第1、3、5、7个月取L3-4、L4-5椎间盘。研究采用胶原VG特染,光、电镜观察及免疫组织化学PAP等方法。结果Sham-0组各阶段纤维环中胶原纤维结构排列规则,偶尔见到较小的串珠状SP阳性反应物。OVX组纤维环中胶原结构排列紊乱、断裂,以后部改变较明显,软骨细胞退变。椎间盘软骨及周围组织可见较多的SP免疫反应阳性神经末梢。结论OVX后椎间盘较早出现退行性改变,结构及功能的完整性破坏;SP的增加与局部炎症反应相关,可能与骨质疏松性腰背疼痛有关。
Objective To study the change of disc structure and substance P distribution in ovariectomized rats and to explore the causes of osteoporotic low back pain. Methods Sixty-four three-month-old female Sprague-Dawley rats were divided into two groups: OVX and Sham-O; the L3-4 and L4-5 intervertebral discs . Study using collagen VG dye, light, electron microscopy and immunohistochemistry PAP and other methods. Results The collagen fibers in the fibrous rings in each stage of Sham-0 group were arranged regularly, and occasionally smaller beaded SP positive reactants were observed. Collagen structure in the annulus fibrosus was disordered and ruptured in OVX group, and the change in the posterior part was obvious. The chondrocytes degenerated. Disc cartilage and surrounding tissue can be seen more SP immune-positive nerve endings. Conclusions The degeneration of the intervertebral disc after OVX was earlier than that of the OVX. The integrity of the structure and function were destroyed. The increase of SP was related to the local inflammatory reaction, which may be related to the osteoporotic low back pain.
