目的探讨肥胖倾向(DIO)、肥胖抵抗(DIO-R)和对照小鼠性激素变化及肥胖引起生殖功能低下的关系。方法分别在小鼠高脂膳食第8、19周各处死小鼠36只,采集血清,分离睾丸、附睾和精囊腺并称重,采用酶联免疫吸附方法检测其血中的性激素水平。结果实验第19周DIO小鼠睾丸系数和附睾系数分别为(0.62±0.08)、(0.25±0.05)g/100 g,明显低于对照小鼠(0.75±0.07)、(0.30±0.02)g/100 g(P<0.01);DIO小鼠的精子活动度(31.37±13.86)%明显低于对照小鼠(49.76±21.71)%;实验第8周DIO小鼠血清促性腺激素释放激素(GnRH)水平(17.82±1.98)mU/mL明显低于对照小鼠(33.46±4.54)mU/mL(P<0.05);第19周DIO小鼠GnRH水平(48.74±8.95)mU/mL明显高于对照组(27.12±54.54)mU/mL(P<0.05)。结论长期肥胖可能影响小鼠生殖器官的发育和精液品质;肥胖小鼠血清性激素水平变化可能与肥胖引起雄性生殖功能下降有一定关系。
Objective To investigate the relationship between obesity tendencies (DIO), obesity resistance (DIO-R) and sex hormone in control mice and reproductive dysfunction caused by obesity. Methods Thirty-six mice were sacrificed on the 8th and 19th weeks of the high fat diet respectively. Serum was collected and the testis, epididymis and seminal vesicle were isolated and weighed. The levels of sex hormones in the blood were detected by enzyme-linked immunosorbent assay. Results At week 19, testicular coefficient and epididymis index of DIO mice were (0.62 ± 0.08) and (0.25 ± 0.05) g / 100 g, respectively, significantly lower than that of control mice (0.75 ± 0.07) and (0.30 ± 0.02) g / (P <0.01). The sperm motility (31.37 ± 13.86)% in DIO mice was significantly lower than that in control mice (49.76 ± 21.71)%. Serum gonadotropin-releasing hormone (GnRH) (17.82 ± 1.98) mU / mL was significantly lower than that of control mice (33.46 ± 4.54) mU / mL (P <0.05). The GnRH level in DIO mice at week 19 (48.74 ± 8.95) mU / mL was significantly higher than that of the control (27.12 ± 54.54) mU / mL (P <0.05). Conclusion Long-term obesity may affect reproductive organs development and semen quality in mice. The changes of serum sex hormone levels in obese mice may be related to the decline of male reproductive function induced by obesity.
