目的研究邻苯二甲酸二(2-乙基己基)酯(DEHP)对雌性小鼠子宫内膜雌激素受体(ER)α表达的影响。方法将健康成年清洁级雌性ICR小鼠60只随机分为4组,即对照(大豆油)组和低(125 mg/kg)、中(500 mg/kg)、高剂量(2 000 mg/kg)DEHP染毒组,每组15只。于早晨采用灌胃方式进行染毒,染毒容量为5 ml/kg,每天1次,每周6 d,连续染毒16周。染毒结束后,于动情间期处死小鼠,采用HE染色法观察子宫组织形态学变化,采用免疫组化染色法检测子宫内膜ERα的表达情况。结果各染毒组小鼠子宫组织固有层腺体数目减少,子宫内膜出现胞核假复层现象。与对照组比较,中、高剂量组子宫内膜免疫组化染色程度较强,范围较大。与高剂量DEHP染毒组比较,对照组和低、中剂量DEHP染毒组小鼠子宫内膜中平均光密度(mean density,MOD)值较高,差异均有统计学意义(P<0.05)。与对照组和低剂量DEHP染毒组比较,中、高剂量DEHP染毒组小鼠子宫内膜中积分光密度(integral optical densitiesI,OD)值较高,差异均有统计学意义(P<0.05)。且随着DEHP染毒剂量的升高,小鼠子宫内膜中MOD值和IOD值均呈上升趋势。结论 DEHP可促进小鼠子宫内膜ERα的表达。
Objective To study the effect of DEHP on estrogen receptor (ER) α expression in the endometrium of female mice. Methods Sixty healthy adult female ICR mice were randomly divided into 4 groups: control (soybean oil) group and low (125 mg / kg), medium (500 mg / kg) ) DEHP exposure group, 15 rats in each group. In the morning by gavage exposure, exposure capacity of 5 ml / kg, once daily, 6 d per week, continuous exposure to 16 weeks. After the exposure, the mice were sacrificed during the period of estrus, the morphology of uterus was observed by HE staining, and the expression of ERα was detected by immunohistochemistry. Results The numbers of lamina propria of uterine tissue of mice in each exposure group decreased and the phenomenon of nuclear pseudostratified in the endometrium. Compared with the control group, the endometrial immunohistochemical staining of middle and high dose group was stronger and larger. Compared with the high dose DEHP group, the mean density (MOD) value of the endometrium of the mice in the control group and those in the low and medium dose DEHP groups were higher (P <0.05) . Compared with the control group and the low-dose DEHP-treated group, the integral optical densities (OD) values of endometrium in DEHP-treated mice were higher than those in the control group and the low-dose DEHP-treated group, the difference was statistically significant (P <0.05 ). And with the increase of DEHP dose, the MOD value and IOD value of mouse endometrium showed an increasing trend. Conclusion DEHP can promote the expression of ERα in mouse endometrium.
