目的建立小鼠肠道外感染轮状病毒(Rotavirus,RV)动物模型,观察该模型动物发病时的细胞免疫功能状态。方法通过腹腔注射RV,建立小鼠肠道外感染RV的模型。分别在病毒接种后第3天及第7天通过摘除眼球取血,流式细胞仪检测T细胞亚群。同时处死动物,无菌取出肺脏、肝脏和脾脏进行组织病理学检查。设立正常对照组。结果小鼠肠道外感染RV成功(可见到肺脏、肝脏和脾脏非特异性病毒感染性炎性病变)。淋巴细胞亚群测定:实验组CD3、CD4及CD4/CD8比值均较对照组明显降低(P<0.01)。结论RV引起小鼠肠道外脏器肺脏、肝脏和脾脏组织病变的同时,机体免疫功能亦明显降低。
Objective To establish an animal model of rotavirus (RV) infection in mice and observe the cellular immune function of the model animals. Methods The mouse model of RV parenteral infection was established by intraperitoneal injection of RV. Blood was taken from the eyeballs on day 3 and day 7 after virus inoculation, respectively. T cell subsets were detected by flow cytometry. At the same time sacrificed animals, aseptic removal of the lungs, liver and spleen for histopathological examination. Set up a normal control group. Results The mice with parenteral infection were successfully treated with RV (non-specific virus-infected inflammatory lesions of lung, liver and spleen were observed). Lymphocyte subsets determination: The experimental group CD3, CD4 and CD4 / CD8 ratio were significantly lower than the control group (P <0.01). Conclusions RV can cause the pathological changes of lung, liver and spleen in the extraintestinal organs of mice, meanwhile, the immunological function of the body is also obviously decreased.
