目的研究银翘解毒片中绿原酸、连翘苷和甘草酸的含量和测定方法。方法采用反相高效液相色谱法,色谱柱为DiamonsilTMC18(250 mm×4.6 mm,5μm),乙腈-0.1%磷酸水溶液为流动相,梯度洗脱,流速为1.0 mL/min,三波长检测,λ1为326 nm,λ2为228 nm,λ3为252 nm,柱温30℃。结果绿原酸、连翘苷和甘草酸进样量分别在0.395 6~2.769 2μg、0.592 8~4.149 6μg和0.558 3~3.908 0μg范围内呈良好的线性关系,相关系数分别为0.999 1、0.999 8和0.999 9,平均加样回收率分别为100.5%、97.1%和99.6%,RSD分别为1.6%、2.1%和1.2%(n=6)。结论该方法专属性好、准确度高,可用于综合评价银翘解毒片的质量。
Objective To study the content and determination methods of chlorogenic acid, forsythin and glycyrrhizic acid in Yinqiaojiedu tablets. Methods The reversed-phase high performance liquid chromatography was used. The chromatographic column was DiamonsilTM C18 (250 mm×4.6 mm, 5 μm). Acetonitrile-0.1% aqueous solution of phosphoric acid was used as the mobile phase. The gradient elution was performed at a flow rate of 1.0 mL/min. Three-wavelength detection was performed. It is 326 nm, λ2 is 228 nm, λ3 is 252 nm, and the column temperature is 30°C. Results The chlorogenic acid, forsythin and glycyrrhizic acid injections were linear in the range of 0.395 6 to 2.769 2 μg, 0.592 8 to 4.149 6 μg and 0.558 3 to 3.908 0 μg, respectively. The correlation coefficients were 0.999 1, 0.999 8 respectively. And 0.999 9, average sample recovery were 100.5%, 97.1% and 99.6%, RSD were 1.6%, 2.1% and 1.2% (n=6). Conclusion The method has high specificity and accuracy and can be used to comprehensively evaluate the quality of Yinqiaojiedu tablets.
