研究一步层析制备高比活抑肽酶新工艺。方法:将化学改性与修饰壳聚糖体,共价偶联配基牛胰蛋白酶,制成固定化酶为抑肽酶亲和吸附剂,直接对黄牛肺提取液一步层析纯化,再超滤脱盐、浓缩、去热原,冻干即抑肽酶。结果:抑肽酶比活5700kiu/mg,收率22×10~4kiu/kg(牛肺);固定化酶单位活力25950kiu/g(干),酶活性回收率55%,非特异性吸附较小,机械强度高,可反复使用。结论:工艺过程简便,样品比活较高,质量稳定,成本低,适合工业化大生产。
Study of a new high-specific activity of aprotinin chromatography. Methods: Chemically modified chitosan and chitosan were covalently conjugated with alanyltransferase to prepare immobilized enzyme for aprotinin affinity adsorbent. Filter desalination, concentrated, to pyrogen, lyophilized that aprotinin. Results: The specific activity of aprotinin was 5700kiu / mg and the yield was 22 × 10 ~ 4kiu / kg (bovine lung). The activity of immobilized enzyme was 25950kiu / g (dry), the recovery of enzyme activity was 55% High mechanical strength, can be used repeatedly. Conclusion: The process is simple, high specific activity, stable quality, low cost, suitable for large-scale industrial production.
