为探讨环氧乙烷(Eto)对细胞内主要修饰酶DNA甲基化酶活性的影响,对Wistar雄性大鼠采用静式吸入染毒,浓度751.7mg/m3,每天2小时,每周6天,连续染毒9周。对其肝、肾和睾丸DNA进行限制性内切酶同裂酶(MboI和Sau3AI,HpaⅡ和MspⅠ)敏感性试验和Southern印迹杂交分析。结果:大鼠肝、肾和睾丸DNA经HindⅢ、MboⅠ和Sau3AⅠ酶解后呈完全酶解带型,仅见1条杂交带。经HpaⅡ和MspⅠ酶解后呈不完全酶解带型,均见多条杂交带。电泳带型、杂交带条数及放射自显影强度在染毒组和对照组之间差异均无显著性。表明Eto对大鼠肝、肾、睾丸DNA甲基化酶活性无影响,睾丸DNA甲基化程度远低于肝和肾。
To investigate the influence of ethylene oxide (Eto) on DNA methyltransferase activity of major intracellular enzymes, Wistar male rats were treated with inhalation of static inhalation at a concentration of 751.7mg / m3, 2 hours a day, 6 Days, continuous exposure to 9 weeks. Restriction endonuclease (MboI and Sau3AI, HpaII and MspI) susceptibility assays and Southern blot hybridization analyzes were performed on their liver, kidney and testis DNA. RESULTS: The DNA of rat liver, kidney and testis was completely digested with Hind Ⅲ, Mbo Ⅰ and Sau3A Ⅰ, and only one hybridized band was found. After digestion with HpaⅡand MspⅠ, it was incompletely digested, showing multiple hybridization bands. Electrophoresis zone type, the number of hybridization bands and autoradiography intensity in the exposure group and the control group, no significant difference. The results showed that Eto had no effect on DNA methylase activity in liver, kidney and testis, and DNA methylation in testis was much lower than that in liver and kidney.
