目的研究奥沙利铂对人肝癌细胞株SMMC7721生长抑制及诱导凋亡的作用并探讨其机制。方法以不同浓度的奥沙利铂作用于SMMC7721细胞,应用MTT法检测奥沙利铂对SMMC7721细胞生长的抑制作用;流式细胞术检测细胞周期和凋亡;Hoechst33258荧光染色法观察细胞凋亡形态学变化;采用分光光度法检测caspase-3、cas-pase-8、caspase-9的活性。结果奥沙利铂可抑制SMMC7721细胞的生长并具有时间和剂量依赖性,Hoechst33258荧光染色可见典型的细胞凋亡特征,流式细胞仪检测到细胞阻滞于S期和SubG1峰。经过奥沙利铂诱导,caspase-3、caspase-9活性被上调,较对照组明显升高(P<0.05),caspase-8活性未见明显改变(P>0.05)。结论奥沙利铂具有抑制肝癌细胞株SMMC7721增殖及诱导凋亡作用,此作用可能通过激活内源性凋亡途径实现。
Objective To investigate the effect of oxaliplatin on the growth inhibition and apoptosis induction of human hepatocellular carcinoma cell line SMMC7721 and explore its mechanism. Methods The inhibitory effect of oxaliplatin on the growth of SMMC7721 cells was detected by MTT assay at different concentrations of oxaliplatin. The cell cycle and apoptosis were detected by flow cytometry. The morphological changes of apoptosis were observed by Hoechst33258 staining. The changes of caspase-3, cas-pase-8 and caspase-9 were detected by spectrophotometry. Results Oxaliplatin inhibited the growth of SMMC7721 cells in a time and dose-dependent manner. Hoechst33258 staining showed typical apoptotic features. Cell arrest was observed in S phase and SubG1 peak by flow cytometry. After oxaliplatin induction, the caspase-3 and caspase-9 activities were up-regulated compared with the control group (P <0.05) and the activity of caspase-8 was not significantly changed (P> 0.05). Conclusion Oxaliplatin can inhibit the proliferation and induce apoptosis of hepatocellular carcinoma cell line SMMC7721, which may be through the activation of endogenous apoptotic pathway.
