目的探索转录因子7类似物2(TCF7L2)基因多态性与福建汉族人群2型糖尿病(T2DM)的相关性。方法2013年7月至2014年6月,采用1∶1匹配病例对照研究方法,选取福建省5个国家疾病监测点社区人群中2型糖尿病患者298例和对照者298人,应用基质辅助激光解吸附电离飞行时间质谱(MALDI-TOF-MS)分析技术对TCF7L2基因上的10个单核苷酸(SNPs)位点(rs11196205、rs12255372、rs4506565、rs7901695、rs7903146、rs7895340、rs11196218、rs12775879、rs290481、rs6585205)进行基因分型,采用SHEsis软件进行Hardy-Weinberg平衡检验,采用Logistic回归分析各SNPs位点与T2DM的相关性。结果 SNPs位点的基因型分布符合Hardy-Weinberg平衡定律;病例组与对照组TCF7L2基因10个SNP位点的基因型和等位基因频率差异无统计学意义(P>0.05);病例组与对照组各SNP位点分布差异无统计学意义(P>0.05)。结论TCF7L2基因10个位点SNPs与福建汉族人群2型糖尿病未发现存在相关性。
Objective To explore the relationship between TCF7L2 gene polymorphism and type 2 diabetes mellitus (T2DM) in Fujian Han population. Methods From July 2013 to June 2014, 298 patients with type 2 diabetes mellitus and 298 controls were enrolled in the community surveillance of 5 national disease surveillance sites in Fujian Province from January 2013 to June 2014. The patients were treated with matrix-assisted laser Ten single nucleotide (SNPs) sites (rs11196205, rs12255372, rs4506565, rs7901695, rs7903146, rs7895340, rs11196218, rs12775879, rs290481, rs6585205) on the TCF7L2 gene were analyzed by adsorption ionization time of flight mass spectrometry (MALDI-TOF- ) Were genotyped. Hardy-Weinberg equilibrium test was performed with SHEsis software. Logistic regression was used to analyze the association between SNPs and T2DM. Results Genotype distribution of SNPs was in accordance with the Hardy-Weinberg equilibrium law. There was no significant difference in the genotype and allele frequencies of 10 SNP loci in TCF7L2 gene between cases and controls (P> 0.05) There was no significant difference in the distribution of SNPs (P> 0.05). Conclusion There is no correlation between 10 loci of TCF7L2 gene and type 2 diabetes in Fujian Han population.
