目的 研究川芎嗪 (TMP)对由乙酰胆碱 (ACh)或高K+ 引起的离体蟾蜍肾上腺髓质嗜铬细胞儿茶酚胺分泌的抑制作用。方法 用胶原酶 ( 0 5mg/mL)消化蟾蜍肾上腺 ,再用牛血清白蛋白 ( 5mg/mL)终止消化 ,获得离体的蟾蜍肾上腺髓质嗜铬细胞。用乙酰胆碱 ( 2 0 0 μmol L)、高K+ ( 6 5mmol L)、川芎嗪 ( 0 5× 10 - 6 ~ 2× 10 - 6 mmol L)或它们的混合液分别作用于离体嗜铬细胞 ,测定其儿茶酚胺 (CA)的分泌量。结果 川芎嗪对乙酰胆碱或高K+ 引起的分泌活性均有明显的抑制作用。结论 离体蟾蜍肾上腺髓质嗜铬细胞可作为刺激_分泌耦联的模型 ;川芎嗪可能是一种钙离子拮抗剂
Objective To study the inhibitory effect of tetramethylpyrazine (TMP) on the secretion of catecholamines from porcine chromaffin cells isolated from acetylcholine (ACh) or high K+. Methods The adrenal glands were digested with collagenase (0.5 mg/mL) and then digested with bovine serum albumin (5 mg/mL) to obtain ex vivo adrenal medulla chromaffin cells. Acetylcholine (200 μmol L), high K+ (65 mmol L), tetramethylpyrazine (0.5×10 −6 to 2×10 −6 mmol L) or a mixture of them was used to treat isolated chromaffin cells. The catecholamine (CA) secretion was measured. RESULTS: Tetramethylpyrazine had a significant inhibitory effect on acetylcholine or high K+ secretion. Conclusion The exoadrenal medulla chromaffin cells can be used as a model of stimulation-secretion coupling; Ligustrazine may be a calcium antagonist
