To observe the expression and significance of Th17 cells and specific transcription factor retinoid-related orphan nuclear receptor (RORγt) in peripheral blood CD4 + T cells in normal pregnant women and patients with preeclampsia. The experimental group was 25 cases of preeclampsia, 20 cases of normal pregnant women and 20 cases of nonpregnant women. The peripheral blood of the subjects was collected. The expression of Th17 cell-associated cytokines IL-17A, IL-6 and TNF-α was detected by enzyme-linked immunosorbent assay (ELISA). Peripheral blood mononuclear cells (PBMCs) CD4 + T lymphocytes and RT-RCR were used to detect the expression of Th17 cell-specific transcription factor RORγt in CD4 + T cells. Flow cytometry was used to detect the expression of CD4 + IL-17 + T cells proportion. The levels of IL-6 and IL-17A in peripheral blood serum of preeclampsia patients were (31.72 ± 13.34) ng / L and (2.61 ± 1.64) ng / L, respectively, which were significantly higher than those of normal pregnant women (18.00 ± 8.64) ng / L and (11.69 ± 3.68) ng / L respectively, the difference was statistically significant (P <0.05); The expression of RORγt mRNA in subgroup The expression of CD4 + IL-17 + T cells in preeclampsia group was significantly higher than that in normal pregnancy group (P <0.01), the expression of CD4 + IL-17 + T cells in preeclampsia group was (1.83 ± 0.42)%, Which was significantly higher than that of the normal pregnancy group (0.87 ± 0.26)% (P <0.01). Abnormal expression of RORγt mRNA, Th17 cells and Th17 cell related cytokines in peripheral blood CD4 + T cells of preeclampsia patients may play an important role in the pathogenesis of the disease.
