九州虫草Cordyceps kyushuensis 菌丝体粗多糖经酶法结合Sevag法除蛋白、H2O2法脱色、透析、乙醇分部沉淀得到Ck1,Ck1经DE-23柱层析得到多糖Ck1-A。经紫外扫描、聚丙烯酰胺凝胶电泳、Sepharose 4B柱层析等方法检测确定Ck1-A为均一组分。苯酚-硫酸法测得Ck1-A的含糖量为86.1%,硫酸-咔唑法测得Ck1-A含有微量的葡萄糖醛酸。Sepharose 4B柱层析测得Ck1-A的分子量为927 kDa。红外光谱和核磁共振氢谱的结果均显示Ck1-A含有α-型糖苷键。体内实验表明Ck1-A对小鼠肉瘤S180有明显的抑制作用。
Cordyceps kyushuensis mycelium polysaccharides were enzymatically combined with Sevag method to remove protein, H2O2 decolorization, dialysis, ethanol fractionation to obtain Ck1, Ck1 by DE-23 column chromatography to obtain polysaccharide Ck1-A. The components of Ck1-A were determined by UV scanning, polyacrylamide gel electrophoresis, and Sepharose 4B column chromatography. The sugar content of Ck1-A was 86.1% as measured by the phenol-sulfuric acid method, and trace amount of glucuronic acid was detected in the Ck1-A method by the sulfuric acid-carbazole method. The molecular weight of Ck1-A measured by Sepharose 4B column chromatography was 927 kDa. The results of infrared spectroscopy and nuclear magnetic resonance spectroscopy show that Ck1-A contains α-glycoside linkages. In vivo experiments showed that Ck1-A had a significant inhibitory effect on mouse sarcoma S180.
