目的:建立手性固定相拆分安倍生坦的高效液相色谱方法。方法:用多糖衍生化手性固定相,考察了流动相中异丙醇比例、柱温、流速对安倍生坦及其对映体手性拆分的影响。采用Chiralpak AD-H色谱柱(250mm×4.6mm,5μm),以正己烷-异丙醇(17.5∶82.5)为流动相,流速为0.4mL·min-1,检测波长263nm,柱温25℃,拆分安倍生坦及其对映体。结果:Chiralpak AD-H色谱柱能使安倍生坦与对映体达到基线分离,(S)-对映体先于(R)-对映体出峰。结论:本法操作简便,可用于安倍生坦及其对映体的分离和质量控制。
Objective: To establish a high performance liquid chromatographic method for the separation of ambrisentan from chiral stationary phases. Methods: Chiral stationary phases were derivatized with polysaccharides. The effects of isopropanol ratio, column temperature and flow rate on the separation of ambrisentan and its enantiomers were investigated. Chiralpak AD-H column (250 mm × 4.6 mm, 5 μm) was used with a mobile phase of n-hexane-isopropanol (17.5:82.5) at a flow rate of 0.4 mL · min -1 and a detection wavelength of 263 nm. Resolution of Abesentan and its Enantiomer. Results: Chiralpak AD-H chromatographic separation of ambrisentan from the enantiomers was achieved. The (S) -enantiomer was peaked at the (R) -enantiomer. Conclusion: This method is simple and can be used for the separation and quality control of ambrisentan and its enantiomers.
