51只大鼠、15只家犬和4只家猫于实验前24h ip优降宁30~50mg/kg bid,实验时先将动物麻醉,再iv利血平2mg/kg,观察血压变化。主动脉条片40条和离体心耳10例,均取自事先用ip优降宁120mg/kg bid处理的家兔。实验时于水浴中加入利血平,观察主动脉条收缩及心率变化。结果表明预先用单胺氧化酶抑制剂优降宁处理的动物,利血平的降压作用可明显翻转为升压作用;利血平能轻度收缩主动脉条并使离体心耳跳动频率轻度加快。利血平的心血管兴奋作用可被α受体阻断剂酚妥拉明和氯丙嗪所对抗,其增大脉压差作用可被β受体阻断剂心得安所减小。提示升压作用的机制是外周肾上腺素能性质的,本文进一步证明在临床抗高血压药物联合应用中,优降宁与利血平不能大剂量序贯联合应用。
Fifty-one rats, 15 dogs and 4 domestic cats received intraperitoneal injection of 30 to 50 mg / kg bid for 24 h prior to the experiment. Animals were anesthetized and then reserpine 2 mg / kg for observing the change of blood pressure. 40 aortic strips and 10 atrial appendages were obtained from rabbits previously treated with ip ginsenoside 120 mg / kg bid. Experiments in the water bath to join reserpine, observed aortic contraction and heart rate changes. The results showed that pre-treated with monoamine oxidase inhibitor gonadotropin, antihypertensive effect of reserpine can be significantly reversed to boost the role; reserpine mild contraction of the aortic strips and isolated atrial fibrillation mildly faster. The cardiovascular stimulant effect of reserpine can be antagonized by the alpha receptor antagonist phentolamine and chlorpromazine, which increases the pulse pressure difference effect can be reduced by beta-blocker propranolol. Suggesting that the mechanism of the role of hypertension is peripheral adrenergic nature of this article further evidence that the combination of clinical antihypertensive drugs, superior to Ning and reserpine can not be used in large doses sequential combination.
