目的对2014年5月由越南回国的女工所感染的麻疹病毒进行基因特征分析。方法采集病人的急性期咽拭子,通过RT-PCR和基因测序分析N基因碳末端450个核苷酸片段,利用BLASTn对序列进行相似性检索,分析麻疹病毒基因特征,并与参考株及沪191疫苗株进行比对,采用Neighbor-Joining方法构建麻疹核蛋白核苷酸种系进化树。结果获得1段相应样本N基因碳末端序列,上传至Gen Bank(KJ994346),该核苷酸序列与越南病毒株(Viet Nam:Ho Chi Minh City-AB928200)序列同源性100%,与WHO参考株(Manchester.UNK/30.94-AF280803)同一谱系,属于D8型,序列同源性为96%,氨基酸同源性为97%,其中产生5个基酸替换,第79位点发生变异,由异亮氨酸替代为苏氨酸,蛋白质极性发生改变,由疏水性变成亲水性。结论该病例为中山市首次发现的输入性D8型麻疹病毒感染病例。
Objective To analyze the genetic characteristics of the measles virus infected by female workers returning from Vietnam in May 2014. Methods Acute pharyngeal swabs were collected from patients. The 450 nucleotides of the N-terminal N gene were analyzed by RT-PCR and gene sequencing. The similarity of the sequences was analyzed by BLASTn, and the genetic characteristics of the measles virus were analyzed. 191 vaccine strains were compared, and the Neighbor-Joining method was used to construct the phylogenetic tree of measles nucleoprotein nucleotide. Results The N-terminal sequence of N gene was amplified and cloned into Gen Bank (KJ994346). The nucleotide sequence was 100% homologous with the sequence of Viet Nam: Ho Chi Minh City-AB928200. Strain (Manchester.UNK / 30.94-AF280803) belongs to the same lineage and belongs to the D8 type. The homology is 96% and the amino acid homology is 97% Leucine instead of threonine, protein polarity changes, from hydrophobic to hydrophilic. Conclusion This case was the first case of imported D8 measles virus in Zhongshan City.
