目的 探讨生长抑素类似物SMS2 0 1 995 (SMS)是否通过诱导人胆管癌细胞凋亡而抑制肿瘤生长 ,以及与细胞内游离钙的关系。方法 采用人胆管癌细胞系SK ChA 1,应用MMT比色法 ,AnnexinV FITC技术及激光共聚焦显微镜技术等方法进行检测和观察。结果 在SMS作用下SK ChA 1细胞生长受抑制。SMS处理SK ChA 1细胞后 ,AnnexinV标记的凋亡细胞增多。细胞凋亡的同时细胞内游离钙浓度升高。结论 SMS可能通过诱导SK ChA 1细胞 [Ca2 + ]i依赖性细胞凋亡 ,抑制胆管癌生长。
Objective To investigate whether somatostatin analogue SMS2 01995 (SMS) can inhibit tumor growth by inducing human cholangiocarcinoma cell apoptosis and its relationship with intracellular free calcium. Methods The human cholangiocarcinoma cell line SK ChA 1 was detected and observed by MMT assay, Annexin V FITC and laser confocal microscopy. Results The growth of SK ChA 1 cells was inhibited by SMS. After the SK ChA 1 cells were treated with SMS, AnnexinV-labeled apoptotic cells increased. Apoptosis at the same time intracellular free calcium concentration increased. Conclusion SMS may inhibit the growth of cholangiocarcinoma by inducing [Ca2 +] i-dependent apoptosis of SK ChA1 cells.
