目的研究白血病细胞及正常人外周血淋巴细胞中的 B- Raf和 Raf- 1表达水平及激酶活性。方法利用 Western印迹法检测 B- Raf和 Raf- 1表达水平,利用免疫沉淀及 Western印迹法检测 Raf- 1及 B- Raf的激酶活性。结果 Raf- 1在白血病细胞及正常人外周血淋巴细胞中均有表达 ,表达水平基本相似,其激酶活性较低; B- Raf仅在白血病细胞中表达,且在 Jurkat和 K562细胞中的表达水平较高,其激酶活性也较高。结论在白血病细胞中, B- Raf的高表达及活化可能是白血病发病的机制之一。
Objective To investigate the expression levels and kinase activity of B-Raf and Raf-1 in peripheral blood lymphocytes of leukemia cells and normal individuals. Methods The expression of B-Raf and Raf-1 was detected by Western blotting, and the kinase activity of Raf-1 and B-Raf was detected by immunoprecipitation and Western blot. Results Raf-1 was expressed in leukemia cells and normal human peripheral blood lymphocytes. The expression levels of Raf-1 were similar, and their kinase activity was low. B-Raf was expressed only in leukemia cells and expressed in Jurkat and K562 cells. Higher, its kinase activity is also higher. Conclusion The high expression and activation of B-Raf may be one of the mechanisms of leukemia in leukemia cells.
