目的:采用开颅机械闭塞法制备局灶性脑缺血大鼠模型,评价黄七胶囊对该模型的影响。方法:SD雄性大鼠90只,分为假手术组11只和造模组79只,对造模组采用开颅机械闭塞法制备局灶性脑缺血模型,根据神经行为学评分评判造模是否成功。将造模成功的50只大鼠分为模型对照组,黄七胶囊低、中、高剂量组(分别灌胃黄七胶囊0.308g/kg体质量、0.616g/kg体质量、1.232g/kg体质量),血塞通组(灌胃血栓通片0.12g/kg体质量),给药14d。比较各组大鼠神经行为学变化、脑梗死面积、血小板聚集程度等指标。结果:黄七胶囊能改善模型大鼠神经行为学评分,减少脑梗死面积,抑制二磷酸腺苷二钠(ADP)诱导的血小板聚集。结论:黄七胶囊对开颅机械闭塞法制备的局灶性脑缺血模型大鼠局灶性脑缺血有明显的改善作用。
OBJECTIVE: To establish a rat model of focal cerebral ischemia by craniotomy mechanical occlusion and evaluate the effect of Huangqi capsule on this model. Methods: Ninety male Sprague Dawley rats were randomly divided into sham operation group (n = 11) and model group (n = 79). Focal cerebral ischemia model was established by craniotomy mechanical occlusion in model group and neuroethology score whether succeed. The 50 successful rats were divided into model control group, Huangqi capsule low, middle and high dose group (intragastric administration of Huangqi capsule 0.308g / kg body weight, 0.616g / kg body weight, 1.232g / kg Body weight), Xuesaitong group (intragastric administration of thrombosis tablets 0.12g / kg body weight), administration 14d. Neurobehavioral changes, infarct size and platelet aggregation were compared between the two groups. Results: Huangqi Capsule can improve neurobehavioral scores of model rats, decrease infarct size and inhibit platelet aggregation induced by ADP. Conclusion: Huangqi Capsule can obviously improve the focal cerebral ischemia in rats with focal cerebral ischemia induced by craniotomy occlusion.
