亚油酸甲酯具有较强的触杀、产卵抑制和驱避作用。对一株粒毛盘菌产油脂突变株Z60胞外油脂进行了GC-MS分析,其中亚油酸甲酯(C18:2)占总脂肪酸甲酯质量分数的32.7%。以亚油酸甲酯含量为指标,在单因素基础上,采用正交实验对亚油酸甲酯的纯化条件进行了优化,最佳条件为3.0 g粒毛盘菌胞外油脂,6.6 g尿素,16.5 mL甲醇溶液,-5℃包合18 h,亚油酸甲酯纯度为50.1%。进一步利用硝酸银薄层层析法对其进行分离,分离后亚油酸甲酯气相色谱纯度提高到96.7%。
Methyl linoleate has a strong contact, spawning inhibition and repellent effect. GC-MS analysis was conducted on the extracellular fat of a strain of lipid-producing mutant of the genus Mycoplasma, in which methyl linoleate (C18: 2) accounted for 32.7% of the total fatty acid methyl ester mass fraction. Using methyl linoleate as an index, the purification conditions of methyl linoleate were optimized by orthogonal experiment on the basis of single factor. The optimum conditions were as follows: 3.0 g extracellular fat, 6.6 g urea , 16.5 mL of methanol solution. Incubation at -5 ℃ for 18 h, the purity of methyl linoleate was 50.1%. Further separation was performed by silver nitrate thin layer chromatography, and the purity of methyl linolenate was increased to 96.7% after separation.
