目的 :检测再生肝细胞表面增殖细胞核抗原 (PCNA)表达分布状态 ,对肝细胞再生程度作出动态评价。方法 :制备亚急性暴发性肝功能衰竭模型 ,以 PCNA单克隆抗体 ,采用免疫组织化学技术检测。结果 :大白鼠术后不同时期 ,AL T、TB水平呈进行性增高 ,术后 2 4h PCNA阳性细胞百分率进行性下降 ,与对照组相比分别为 2 4h(18.8± 11.2 ) % / (30 .7± 16 .8) % (P <0 .0 5 ) ,36 h (7.7± 5 .7) % / (2 8.5± 12 .3) % (P <0 .0 1) ,48h(5 .7±8.9) % / (2 5 .8± 8.9) % (P <0 .0 1) ,72 h(2 .8± 6 .4) % / (15 .6± 7.2 ) % (P <0 .0 1)。结论 :实验性亚急性暴发性肝功能衰竭模型中肝细胞的再生进行性受到抑制 ,可能与机体免疫因素参与有关。
Objective: To detect the expression and distribution of proliferating cell nuclear antigen (PCNA) on the surface of regenerating hepatocytes and to evaluate the degree of hepatocyte regeneration. Methods: Subacute fulminant hepatic failure model was prepared and detected by PCNA monoclonal antibody using immunohistochemistry. Results: The ALT and TB levels increased progressively at different stages after operation in rats. The percentage of PCNA positive cells decreased progressively at 24 hours after operation, which were respectively 24 h (18.8 ± 11.2)% / (30. 7 ± 16.8%, P <0.05), 36 h (7.7 ± 5 .7)% / (2 8.5 ± 12.3)% (P <0.01), 48 h ± 8.9% / (25.8 ± 8.9)% (P <0.01), 72 h (2.8 ± 6.4%) / (15.6 ± 7.2)% 1). Conclusions: The progress of hepatocyte regeneration in experimental subacute fulminant hepatic failure model is inhibited, which may be related to the involvement of immune factors.
