目的研究脾酪氨酸激酶(spleen tyrosine kinase,Sy K)和肝细胞生长因子(hepatocyte growth factor,HGF)在乳腺癌表达及临床意义。方法采用免疫组织化学方法分别检测120例乳腺癌组织、100例正常乳腺组织、100例纤维腺瘤组织之间的Sy K和HGF蛋白表达,分析乳腺癌组织中二者的表达水平与临床病理学特征的关系。结果 120例乳腺癌组织中Sy K、HGF的表达率分别为30.0%、68.3%,100例正常乳腺组织表达率分别为60%、33%,100例纤维腺瘤组织表达率分别为63%、36%,Sy K、HGF的表达率在乳腺癌组织与其他两组之间比较,差异有统计学意义(P<0.01);而二者表达率在正常乳腺组织与纤维腺瘤组织之间比较,差异无统计学意义(P>0.05)。Sy K、HGF的表达与乳腺癌临床分期、淋巴结转移相关(P<0.05),与年龄、肿块直径及脉管侵犯无关(P>0.05)。随乳腺癌临床分期越晚、有淋巴结转移者,Sy K表达率缺失越明显,HGF的表达水平依次升高,差异有统计学意义(P<0.05)。在乳腺癌中Sy K与HGF之间表达呈负相关(r=-0.649,P<0.01)。结论 Sy K的表达缺失及HGF在乳腺癌的表达增高与乳腺癌的增殖、侵袭和转移等生物学行为密切相关,联合检测Sy K、HGF在乳腺癌组织中的表达情况有助于乳腺癌的早期诊断及预后的判断。
Objective To study the expression of spleen tyrosine kinase (Sy K) and hepatocyte growth factor (HGF) in breast cancer and its clinical significance. Methods The expressions of Sy K and HGF in 120 cases of breast cancer tissues, 100 cases of normal breast tissues and 100 cases of fibroadenoma tissues were detected by immunohistochemistry. The expressions of both Syk and HGF proteins in breast cancer tissues were analyzed with clinicopathological Characteristics of the relationship. Results The expression rates of Sy K and HGF in 120 cases of breast cancer were 30.0% and 68.3% respectively. The expression rates of 100 cases of normal breast tissues were 60% and 33% respectively. The expression rates of 100 cases of fibroadenoma were 63% The expression rates of Sy K and HGF in breast cancer tissues were significantly different from those in the other two groups (P <0.01), while the expression rates of Sy K and HGF were significantly different between normal breast tissues and fibroadenoma tissues , The difference was not statistically significant (P> 0.05). The expression of Sy K and HGF was correlated with the clinical stage and lymph node metastasis of breast cancer (P <0.05), but not with age, mass diameter and vascular invasion (P> 0.05). With later clinical stage of breast cancer, patients with lymph node metastasis had more obvious Sy K expression loss and HGF expression in turn, with statistical significance (P <0.05). The expression of Sy K was negatively correlated with HGF in breast cancer (r = -0.649, P <0.01). Conclusion The lack of expression of Sy K and the increased expression of HGF in breast cancer are closely related to the biological behaviors such as proliferation, invasion and metastasis of breast cancer. The joint detection of the expression of Sy K and HGF in breast cancer contributes to the development of breast cancer Early diagnosis and prognosis judgment.
