目的 :研究外阴癌中p5 3的表达及突变情况 ,以探索 p5 3突变与外阴癌发生的相关性。方法 :采用 p5 3单克隆抗体DO 7检测 p5 3的表达情况 ,采用SSCP银染方法检测p5 3第 5 8外显子突变情况。结果 31例外阴癌中 p5 3阳性表达 16例 ,占 5 1.6 %。SSCP银染方法检测 p5 3突变也有 16例 ,占 5 1.6 % ,但与免疫组化结果并不一致。结论 :外阴癌中有较高的p5 3突变率 ,同时应用免疫组化及SSCP银染方法检测可提高突变型 p5 3的检出率。
Objective: To study the expression and mutation of p5 3 in vulvar cancer, so as to explore the correlation between the mutation of p5 3 and the occurrence of vulvar cancer. Methods: p5 3 monoclonal antibody DO 7 was used to detect the expression of p5 3, and SSCP silver staining was used to detect the mutation of exon 58 of p5 3. Results The positive expression of p5 3 in 31 cases of vulvar cancer was 16 cases (5 1.6%). SSCP silver staining method detected p5 3 mutation also 16 cases, accounting for 5 1.6%, but not consistent with the results of immunohistochemistry. Conclusion: There is a high mutation rate of p5 3 in vulvar cancer, and the detection rate of mutant p5 3 can be increased by immunohistochemistry and SSCP silver staining.
