目的 测定人膀胱移行细胞癌组织中 ,不同病理分期的人细胞 5型腺病毒早期转录单位E1A因子 (E1AF)和基质金属蛋白酶 2 (MMP 2 )的表达 ,探讨二者之间关系及其临床意义。 方法 采用RT PCR和WesternBlot法 ,检测 41例人膀胱癌组织中E1AF和MMP 2的mRNA及蛋白表达 ,并运用计算机图像扫描分析仪测定图像的吸光度值 (A值 )进行半定量分析。 结果 2 6例表浅性膀胱癌中 ,E1AF的阳性率为 2 6 90 % ,MMP 2和E1AF的A值分别为 36 0 9± 5 82和 2 9 0 8± 6 2 6 ;15例浸润性膀胱癌中 ,E1AF的阳性率为 6 6 6 7% ,MMP 2和E1AF的A值分别为 119 98± 10 44和 89 6 2±10 96。E1AF的阳性率、MMP 2和E1AF的A值在两组均有显著性差异 ;E1AF在G3级膀胱癌组织中的表达量显著高于G1级 ;MMP 2和E1AF的表达呈正相关 (r=0 5 36 ,P <0 0 1)。 结论 E1AF调节MMP 2的表达 ;二者高表达与膀胱癌浸润有关
Objective To determine the expression of E1AF and MMP2 in human bladder adenocarcinoma of human bladder transitional cell carcinoma of different stages with different pathological stages and to explore their relationship and their clinical significance . Methods The mRNA and protein expressions of E1AF and MMP-2 in 41 human bladder cancer tissues were detected by RT-PCR and Western Blot. The absorbance values (A values) of the images were analyzed semi-quantitatively by computer image scanning analyzer. Results In 26 cases of superficial bladder cancer, the positive rate of E1AF was 26.9%, the A values of MMP 2 and E1AF were 36 0 9 ± 5 82 and 29 08 ± 6 26, respectively. Fifteen infiltrative In bladder cancer, the positive rate of E1AF was 66.67%, and the A values of MMP 2 and E1AF were 119 98 ± 10 44 and 89 6 2 ± 10 96, respectively. The positive rate of E1AF, the value of MMP 2 and the value of E1AF in both groups were significantly different. The expression level of E1AF in G3 bladder cancer was significantly higher than that in G1. The expression of MMP 2 and E1AF was positively correlated (r = 0 5 36, P <0 0 1). Conclusion E1AF regulates the expression of MMP-2, which is related to the infiltration of bladder cancer
