目的:观察右归胶囊防治去卵巢大鼠骨质疏松的作用,并探讨其相关的作用机制。方法:SPF级SD大鼠105只,其中15只大鼠作为假手术组,其余大鼠采用去卵巢大鼠复制骨质疏松模型,取造模后雌性大鼠随机分为模型组,右归胶囊低、中、高剂量组(0.71,1.43,2.85 g·kg~(-1)),雌激素组(己烯雌酚,0.02 mg·kg~(-1))和仙灵骨葆胶囊组(0.25 g·kg~(-1)),每组15只,于术后第7天开始ig给药,连续给药6个月。于末次给药后24 h大鼠眼眶采血,测定血清钙(Ca),磷(P),碱性磷酸酶(ALP),酸性磷酸酶(ACP)。完整取出大鼠左右股骨,用双能X射线吸收法骨密度仪测大鼠左股骨骨密度(BMD)和骨矿质(BMC)。用WDW型电子万能实验机测大鼠右股骨力学性能指标挠度、最大力和抗弯强度。结果:与假手术组比较,模型组大鼠血清Ca,P,ALP及ACP水平明显升高,大鼠BMD及BMC含量明显降低,大鼠右股骨挠度、最大力和抗弯强度明显降低(P<0.05,P<0.01);与模型组比较,右归胶囊低、中、高剂量组均能明显降低去势大鼠血清Ca,P,ALP及ACP水平,明显提高去势大鼠BMD及BMC含量,明显升高右股骨挠度、最大力和抗弯强度(P<0.05,P<0.01)。结论:右归胶囊能改善去势大鼠生物力学指标,并能够延缓大鼠骨量丢失,提示其具有预防骨质疏松症作用。
Objective: To observe the effect of Yougui capsule on prevention and treatment of osteoporosis in ovariectomized rats and to explore its related mechanism. Methods: 105 Sprague-Dawley rats were randomly divided into sham-operation group (15 rats) and osteoporosis rats (ovariectomized rats). The rats were randomly divided into model group (0.71, 1.43, 2.85 g · kg -1), estrogen group (diethylstibestrol, 0.02 mg · kg -1) and Xianling Gubao capsule group (0.25 g · kg -1) kg ~ (-1)), 15 in each group. The animals were given ig administration on the 7th day after the operation and continuously administered for 6 months. Blood was collected from the orbit of rats 24 h after the last administration, and serum Ca, P, ALP and ACP were measured. Left and right femurs were completely removed and bone mineral density (BMD) and bone mineral (BMC) of left femur were measured by dual energy X-ray absorptiometry. The mechanical properties of right femur, such as deflection, maximum force and flexural strength, were measured by WDW electronic universal testing machine. Results: Compared with the sham operation group, the levels of Ca, P, ALP and ACP in the model group were significantly increased, while the content of BMD and BMC in the model group was significantly decreased. The right femur flexion, maximum force and flexural strength were significantly decreased (P <0.05, P <0.01). Compared with the model group, Yougui capsule low, medium and high dose groups can significantly reduce the serum Ca, P, ALP and ACP levels in ovariectomized rats, and significantly increase the BMD and BMC Content, significantly increased right femur deflection, maximum force and flexural strength (P <0.05, P <0.01). CONCLUSION: Yougui capsule can improve the biomechanical parameters of ovariectomized rats and delay the loss of bone mass in rats, suggesting that it can prevent osteoporosis.
