目的确定嗜铬颗粒素C/分泌颗粒素Ⅱ(CgC/SgⅡ)和胰抑素(PS)在胰腺导管癌细胞中的表达.方法采用荧光双标记免疫组织化学技术结合激光扫描共聚焦显微镜观察胰腺导管癌(1996/1997,n=10)细胞中CgC/SgⅡ和PS的表达.结果在CgC/SgⅡ单克隆抗体染色后发现,胰腺导管癌的大多数癌细胞呈CgC/SgⅡ免疫反应阳性,免疫反应阳性物质位于细胞胞质内,而细胞核为阴性.CgC/SgⅡ免疫阳性物质在胞质内为均质性或颗粒状,在一些腺管的癌细胞内存在较大的颗粒状荧光物质.同时用PS抗体的染色发现,在高分化胰腺导管癌CgC/SgⅡ免疫阳性的细胞大部分同时表达PS,但在中分化胰腺导管癌CgC/SgⅡ免疫阳性的细胞仅有一部分细胞表达PS.而且PS阳性物质在高分化胰腺导管癌细胞多为均质性,而在中分化胰腺导管癌细胞为均质性和颗粒状荧光,一些细胞的颗粒状荧光物质仅呈CgC/SgⅡ免疫阳性.结论胰腺导管癌细胞中存在嗜铬颗粒素C和胰抑素.
Objective To determine the expression of chromogranin C/secretase II (CgC/SgII) and pancreatic trypsin (PS) in pancreatic ductal carcinoma cells. Methods The expression of CgC/Sg II and PS in pancreatic ductal carcinoma (1996/1997, n=10) was observed by fluorescence double labeling immunohistochemical technique combined with laser scanning confocal microscopy. Results After CgC/SgII monoclonal antibody staining, most of the pancreatic ductal carcinoma cells showed positive CgC/SgII immunoreactivity. The immunoreactive positive substance was located in the cytoplasm and the cell nucleus was negative. CgC/SgII immunopositive substances are homogeneous or granular in the cytoplasm, and there are large granular fluorescent substances in the cancer cells of some glandular ducts. At the same time, staining with PS antibody revealed that most of the highly differentiated pancreatic ductal carcinoma CgC/SgII immunopositive cells express PS at the same time, but only a part of the cells expressed PS in the moderately differentiated pancreatic ductal carcinoma CgC/SgII immunopositive cells. Moreover, PS-positive substances were more homogeneous in well-differentiated pancreatic ductal carcinoma cells, while in the moderately differentiated pancreatic ductal carcinoma cells were homogeneous and granular fluorescence, and some cells had only granular CgC/SgII immunoreactivities. 4. Conclusion There is chromogranin C and Dysstatin in pancreatic ductal carcinoma cells.
