目的:研究乳康汤防治乳腺增生病的作用机制。方法:采用雌-孕激素联合造模形成乳腺增生大鼠模型,以乳结平作阳性对照,观察3个不同剂量乳康汤组大鼠乳头直径、高度和形态学变化,测定模型大鼠血清雌二醇(E2)、泌乳素(PRL)、孕酮(P)含量和雌激素受体(ER)、孕激素受体(PR)阳性表达强度,检测模型大鼠细胞调亡比例。结果:乳康汤可使乳腺增生模型大鼠乳头红肿或增生减轻,明显减少乳腺小叶腺泡数及腺导管上皮细胞层数,可降低模型大鼠血清雌二醇(E2)、泌乳素(PRL)而升高孕酮(P)含量,对乳腺组织中ER、PR的阳性表达强度明显抑制,促进细胞凋亡。结论:乳康汤可通过调节乳腺增生大鼠的激素水平,改善机体内分泌环境,使其对雌激素的敏感性下降,对大鼠乳腺增生有明显的抑制作用。
Objective: To study the mechanism of Rukang Decoction in preventing and treating mammary gland hyperplasia. Methods: The model of hyperplasia of mammary glands was established by estrogen-progestin and the positive control of lactation was taken as the positive control. The diameter, height and morphology of the nipple of 3 different doses of Rukangtang were observed. The serum estradiol (E2), prolactin (PRL), progesterone (P) and estrogen receptor (ER), progesterone receptor (PR) positive expression intensity of the model of rat apoptosis ratio. Results: Rukang decoction can make the nipple swelling or hyperplasia of mammary gland hyperplasia model rat reduce, significantly reduce the number of acinar lobes and the number of glandular duct epithelial cells, can reduce the model rats serum estradiol (E2), prolactin (PRL ) Increased the content of progesterone (P), and significantly inhibited the expression of ER and PR in mammary gland tissue, and promoted the apoptosis. Conclusion: Rukang Decoction can regulate the hormone level of mammary gland hyperplasia rats, improve the endocrine environment of the rats, decrease the sensitivity to estrogen and inhibit the proliferation of mammary glands in rats.
