目的:检测哮喘小鼠小气道及肺组织IL-21、MMP-2与TIMP-1的表达及布地奈德干预对其表达的影响;分析IL-21与MMP-2、TIMP-1及MMP-2/TIMP-1比值的相关关系。方法:建立哮喘小鼠模型。36只雌性小鼠随机分为3组,每组12只。分别为生理盐水对照组(CTRL组)、卵清蛋白(ovalbumin,OVA)致敏、激发哮喘组(OVA组)、布地奈德干预组(OVA+BUD组)。观察各组小气道及肺组织HE及Masson染色情况;免疫组化检测MMP-2、TIMP-1蛋白的表达;RT-PCR检测IL-21的表达。结果:HE及Masson染色结果示哮喘小鼠模型制作成功;OVA组MMP-2与TIMP-1的表达、MMP-2/TIMP-1比值高于CTRL组与OVA+BUD组,其差异有统计学意义(P<0.05);CTRL组与OVA+BUD组比较,其差异无统计学意义(P>0.05);OVA组IL-21的表达低于CTRL组与OVA+BUD组,其差异有统计学意义(P<0.05);CTRL组高于OVA+BUD组,其差异有统计学意义(P<0.05)。结论:OVA组IL-21的表达低于CTRL组和OVA+BUD组;OVA组MMP-2、TIMP-1的表达及MMP-2/TIMP-1比值高于CTRL组和OVA+BUD组。布地奈德干预后,IL-21表达上调,MMP-2、TIMP-1的表达与MMP-2/TIMP-1比值降低IL-21与MMP-2、TIMP-1的表达及MMP-2/TIMP-1比值呈负相关。
Objective: To detect the expression of IL-21, MMP-2 and TIMP-1 in small airway and lung tissue of asthmatic mice and the effect of budesonide intervention on the expression of IL-21, MMP-2, TIMP- 2 / TIMP-1 ratio of the correlation. Methods: To establish a mouse model of asthma. Thirty-six female mice were randomly divided into three groups of 12 rats. The rats in control group (CTRL group), ovalbumin (OVA) group, asthmatic group (OVA group) and budesonide intervention group (OVA + BUD group) were sensitized respectively. The small airway and lung tissue HE staining and Masson staining were observed. The expressions of MMP-2 and TIMP-1 protein were detected by immunohistochemistry. The expression of IL-21 was detected by RT-PCR. Results: The results of HE and Masson staining showed that the asthma model was successfully established. The expression of MMP-2 and TIMP-1 in OVA group was higher than that in CTRL group and OVA + BUD group, the difference was statistically significant (P <0.05). There was no significant difference between CTRL group and OVA + BUD group (P> 0.05). The expression of IL-21 in OVA group was lower than that in CTRL group and OVA + BUD group (P <0.05). There was a significant difference between CTRL group and OVA + BUD group (P <0.05). Conclusion: The expression of IL-21 in OVA group was lower than that in CTRL group and OVA + BUD group. The expression of MMP-2 and TIMP-1 and the ratio of MMP-2 / TIMP-1 in OVA group were higher than those in CTRL group and OVA + BUD group. After the intervention of budesonide, the expression of IL-21 was up-regulated. The ratio of MMP-2, TIMP-1 and MMP-2 / TIMP-1 decreased the expression of IL-21, MMP-2 and TIMP- -1 ratio was negatively correlated.
