采用HPLC-DAD法,同时测定六味五灵片中连翘酯苷A,特女贞苷,连翘苷,五味子醇甲,五味子酯甲,五味子甲素和五味子乙素共7种有效成分的含量。采用Inertsil~?ODS-3色谱柱(250 mm×4.6 mm,5μm),流动相为0.1%磷酸水(A)-乙腈(B),梯度洗脱,柱温25°C,检测波长230 nm。7种化合物在相应的浓度范围内线性关系良好,相关系数均大于0.999。检测限与定量限分别在0.03-0.28μg/m L和0.11-0.87μg/m L之间。精密度、稳定性与重复性的RSD均小于2.34%,平均回收率均在96.71%-103.9%之间。建立的含量测定方法可有效对山东世博金都医药有限公司的十批样品进行检测,为六味五灵片的质量控制与下一步药理研究提供科学依据。
HPLC-DAD method was used to determine the content of seven effective components of forsythiaside A, procyanidin, forsythin, schizandrin, schisandrin, schisandrin and schisandrin in Liuwei Wuling tablet . The mobile phase consisted of 0.1% phosphoric acid water (A) -acetonitrile (B) with gradient elution at a column temperature of 25 ° C with a detection wavelength of 230 nm using an Inertsil® ODS-3 column (250 mm × 4.6 mm, 5 μm). The seven compounds showed good linearity in the corresponding concentration range with the correlation coefficients greater than 0.999. The limits of detection and quantification were between 0.03-0.28μg / m L and 0.11-0.87μg / m L, respectively. The RSDs of precision, stability and repeatability were both less than 2.34% and the average recoveries were between 96.71% and 103.9%. The established method for determination of content can effectively test ten batches of samples of Shandong Expo Jindu Pharmaceutical Co., Ltd. and provide a scientific basis for the quality control and the next pharmacological study of Liuwei Five Ling tablets.
