目的:探究Dicer在胃癌中的表达情况、与临床病理参数的相关性及对细胞生长和迁移的作用。方法:收集56例胃癌患者的癌组织样本和癌旁组织样本,利用实时定量PCR的方法检测Dicer在这些样本中的表达情况。选用2株人胃癌细胞系AGS和MGC803,RNAi技术干扰下调Dicer的表达,CCK-8法分析Dicer表达下调对胃癌细胞生长情况的影响,迁移小室实验分析Dicer表达下调对胃癌细胞体外迁移能力的影响。结果:56例胃癌患者中,14例(25.0%)患者Dicer表达水平升高,27例(48.2%)患者表达降低,15例(26.8%)患者表达无明显变化;Dicer mRNA的表达降低与胃癌患者淋巴结转移和TNM分期存在关联,而与患者性别、年龄、肿瘤大小及癌细胞分化程度无关。沉默Dicer表达后,两株胃癌细胞的生长和迁移能力显著升高。结论:Dicer在胃癌组织中表达降低,且与患者淋巴结转移和TNM分期相关,其低表达有利于胃癌细胞的恶性生长和侵袭转移。
Objective: To investigate the expression of Dicer in gastric cancer, the correlation with clinicopathological parameters and its effect on cell growth and migration. Methods: Tissues from 56 cases of gastric cancer and adjacent normal tissues were collected. The expression of Dicer in these samples was detected by real-time quantitative PCR. Two human gastric cancer cell lines AGS and MGC803 were selected and RNAi technology was used to down-regulate the expression of Dicer. The effect of down-regulation of Dicer expression on the growth of gastric cancer cells was analyzed by CCK-8. The migration chamber assay was used to analyze the effect of Dicer expression down-regulation on gastric cancer cell migration in vitro . Results: Dicer expression increased in 14 cases (25.0%) of 56 gastric cancer patients, decreased in 27 cases (48.2%) and in 15 cases (26.8%), while decreased expression of Dicer mRNA in gastric cancer Patients with lymph node metastasis and TNM staging correlated, but not with the patient’s gender, age, tumor size and cancer cell differentiation degree. After silencing Dicer expression, the ability of both gastric cancer cells to grow and migrate significantly increased. Conclusion: The expression of Dicer in gastric cancer is decreased, and it is correlated with lymph node metastasis and TNM stage. The low expression of Dicer is beneficial to the malignant growth and invasion and metastasis of gastric cancer.
