目的探讨乙酸铅对小鼠睾丸细胞活性和活性氧(ROS)水平的影响。方法将具有小鼠原代睾丸支持细胞特性的TM4细胞株暴露于不同浓度乙酸铅溶液中24 h,采用新型四唑盐WST-8方法检测乙酸铅诱导的细胞增殖情况;采用流式细胞术检测细胞内ROS水平变化。结果与对照组相比,低浓度铅诱导细胞增殖,随着乙酸铅浓度升高,细胞存活率逐渐降低(P<0.05),呈现先增高后降低的剂量-效应关系。ROS结果显示在一定剂量范围内,随着乙酸铅浓度的升高,细胞内ROS含量显著升高(P<0.05)。结论低剂量乙酸铅对小鼠睾丸细胞有兴奋效应,高剂量会抑制细胞增殖,同时可诱导氧化应激反应,引起细胞损伤。
Objective To investigate the effect of lead acetate on testicular cell activity and reactive oxygen species (ROS) in mice. Methods TM4 cell lines with mouse primary testicular supportive cells were exposed to different concentrations of lead acetate solution for 24 hours. The new tetrazolium salt WST-8 method was used to detect lead acetate-induced cell proliferation. Flow cytometry Intracellular ROS levels change. Results Compared with the control group, the cell proliferation was induced by low concentration of lead. With the increase of lead acetate concentration, the cell survival rate decreased gradually (P <0.05), and the dose-response relationship first increased and then decreased. ROS results showed that within a certain dose range, with the lead concentration increased, intracellular ROS content was significantly increased (P <0.05). Conclusion Low-dose lead acetate can stimulate the testicular cells in mice. High doses can inhibit cell proliferation and induce oxidative stress and cause cell damage.
