目的探讨摄入某品牌54°食用白酒后,不同时长及不同剂量对大鼠血糖及胰岛A细胞表达胰高血糖素(Glu)的影响,以期为科学健康饮酒提供一定的基础实验依据。方法正常成年雄性SD大鼠69只,分为正常对照组(15只)及实验组(54只)。其中实验组分为4周组、8周组、12周组;各周组又分为低剂量组[0.8 m L/(kg·d)]、中剂量组[1.6 mL/(kg·d)]、高剂量组[2.4 mL/(kg·d)]。实验组大鼠每天11:00及17:00各灌胃1次,正常对照组不予处理。分别于第4、8、12周末测空腹血糖,取胰尾组织。采用免疫组化SABC法、图像分析及形态计量法进行研究。结果 (1)各实验组大鼠空腹血糖水平与正常对照组比较,差异无统计学意义(P>0.05)。(2)免疫组化结果显示:光镜下,Glu免疫反应阳性产物呈棕黄至棕褐色细颗粒状,位于胞质内。与正常对照组比较,除12周低剂量组稍有升高(P<0.05)外,余实验各组Glu阳性细胞的免疫染色强度及平均光密度的差异无统计学意义(P>0.05)。(3)形态学计量结果显示:与正常对照组比较,各实验组Glu阳性细胞的面数密度均未见明显变化(P>0.05)。结论在本实验设定的剂量和时程内用该白酒灌胃,大鼠血糖未见明显异常;对胰岛A细胞的表达和N_A无明显影响。
Objective To investigate the effect of different doses and doses of long-term intake of certain brands of 54 ° edible white wine on blood glucose and glucagon (Glu) expression in rat islet A cells in order to provide some basic experimental evidence for scientific and healthy drinking. Methods Sixty-nine male SD rats were divided into normal control group (n = 15) and experimental group (n = 54). The experimental group was divided into four groups, eight-week group and twelve-week group. Each week group was divided into low-dose group [0.8 m L / (kg · d)] and middle dose group [1.6 mL / (kg · d) ], High-dose group [2.4 mL / (kg · d)]. Rats in the experimental group were given gavage at 11:00 and 17:00 daily, while the normal control group was not treated. Fasting blood glucose was measured at the end of the 4th, 8th and 12th week respectively, and the tail tissue was taken. Immunohistochemical SABC method, image analysis and morphometry were used to study. Results (1) There was no significant difference in fasting blood glucose between experimental groups and normal control group (P> 0.05). (2) The results of immunohistochemistry showed that under the light microscope, the products of Glu immunoreactivity were fine brown to tan, located in the cytoplasm. Compared with the normal control group, there was no significant difference in the intensity of immunostaining and the average optical density of Glu positive cells except for the 12-week low-dose group (P <0.05). (3) Morphometric results showed that there was no significant change in the number density of Glu positive cells in each experimental group compared with the normal control group (P> 0.05). Conclusion In the experiment set the dose and duration of the liquor with gavage, no significant abnormal blood glucose in rats; the expression of islet A cells and N_A no significant effect.
