目的观察槲皮素对巨噬细胞泡沫化和三磷酸腺苷结合盒式转运体Al(ABCAl)表达的影响,探讨槲皮素抗动脉粥样硬化机制。方法体外培养RAW264.7细胞,与l00mg/L氧化低密度脂蛋白(ox-LDL)共孵育48h建立泡沫细胞模型,不同浓度(20、40和80μmol/L)槲皮素预处理进行干预,油红O染色检测细胞内脂质蓄积情况,免疫荧光及免疫印迹法检测ABCAl蛋白的表达变化。结果模型组细胞内红色脂滴明显增多,给予槲皮素后细胞内脂滴减少,以40、80μmol/L槲皮素作用更为明显;模型组荧光较对照组增强,随槲皮素剂量增加,荧光加进一步增强;模型组ABCAl表达较对照组增加,而槲皮素预处理后,进一步上调ABCAl表达,且其表达强度随槲皮素作用浓度的增加而增强。结论槲皮素能上调ABCAl的表达,抑制巨噬细胞源性泡沫细胞形成。
Objective To investigate the effects of quercetin on the foaming of macrophages and the expression of adenosine triphosphate (ATP) cassette transporter Al (ABCA1), and to explore the anti-atherosclerotic mechanism of quercetin. Methods RAW264.7 cells were cultured in vitro and incubated with ox-LDL (l00mg / L) for 48h to establish a foam cell model. The cells were pretreated with different concentrations of quercetin (20, 40 and 80μmol / L) Red O staining detection of intracellular lipid accumulation, immunofluorescence and Western blot detection ABCAl protein expression changes. Results In the model group, the number of red lipid droplets in the model group was significantly increased. The lipid droplets in the cells decreased after the administration of quercetin, and the effect of quercetin at 40 and 80μmol / L was more obvious. Compared with the control group, , And the fluorescence was further enhanced. The expression of ABCA1 in the model group increased compared with the control group, and after quercetin pretreatment, ABCA1 expression was further up-regulated, and its expression intensity increased with the increase of the concentration of quercetin. Conclusion Quercetin can up-regulate the expression of ABCA1 and inhibit the formation of macrophage-derived foam cells.
